Automated Cell Pellet Resuspension from Large-Volume Reservoir Plates Using a Hamilton VANTAGE

Has anyone successfully automated the centrifugation and resuspension of cells in a reservoir plate or other large-volume, automation-compatible plate format?

I am developing a workflow using a Hamilton VANTAGE and a HighRes MicroSpin. The sample volumes may range from approximately 5 mL to 50 mL per sample, so standard 96-well deep-well plates may not be suitable. Ideally, I am looking for a plate or reservoir with a small number of large-volume wells lke a 4 well reservoir (I am current looking at the Agilent 4 Volumn Reservoir (201308-100). It is separated in pyramid base shapes at the bottom which I think would be allow me to have exact places where to position the pipette tip for re-suspension. I was also thinking about a V bottom and having the Hamilton Vantage dispense while moving in the well, but I don’t know if that is possible.

Resuspension of magentic bead pellets is somewhat similar. There I found that dry shaking improved the resuspension - so after liquid removal go full blast on an HHS or similar.

You can also move the tips to fixed locations in Venus software. This is not controlled by Venus and thus it can lead to crashes. I am however not sure, if that works in liquid handling. Another way would be, to use several virtual labwares with slightly adjusted positions and iterate over them.

My “worst case scenario” was to do this. Just have different labwares with different positions for each well. I was trying to see if there was a way to move while dispensing, but I don’t know if that is even possible.

This is interesting. The currently manual method does spin down in a 50 mL tube. I will look into this. Thank you for the recommendation!!!
