We’ve had some success using our easypick method, but are still running into some challenges. We plate onto an sbs single well agar plate using the core head, and have 96 regions/wells on the plate.
- When we plate the colonies, we plate multiple dilutions (Neat, 1:10, 1:100, 1:1000) to account for varying colony density. In some cases, the colony density might be too high for a few wells in one dilution, so we pick from the more dilute/less dense colony plate. I was trying to think of a way to accommodate this within a single run without putting the burden on the user to omit wells that were picked from the previous agar plate when selecting pickable colonies. Having trouble coming up with anything given that it seems like the easypick software only really allows you to analyze the entire plate using a predefined labware config.
- Honing in on the proper camera settings continues to be a challenge. I think I’ve got a pretty good grip on the colony settings, but can’t get a good enough quality image to get good enough colony selection resolution. Any tips/templates would be appreciated here.
