ELISAs with 8-channel FCA

Hello!

Has anyone created a successful ELISA script with an 8-channel pipetting arm in Fluent Control? I’m currently experiencing significantly higher capsid titer results with the Tecan, and I’m wondering if this is due to the duration of time it takes to wash the plate with an 8-channel arm (~15 minutes for 4 washes). Alternatively, I’m not sure if this is being caused by not manually tapping out residual liquid after washing as I would usually do in a human-run ELISA. Thanks!

There are numerous reasons why the titer results could be higher,

plate washing could definitely lead to higher signals/results - especially if you’re using a pipetting arm as a washer, 1 x column at a time

how is the assay background when compared to manual assay or tapping out ?

can you run the assay with offline washing ?

did you qualify the tecan pipetting for samples/dilutions etc?

i’ve seen that the Tecan usually needs some qualification/optimization to best yield the correct volumes across the pipetting range for the assay
