Serum Liquid Class Settings issue

Hi,

I am currently trying to make a liquid class using the LVK with the aim of aspirating and dispensing Serum multiple times using the same set of tips.
Currently i have to use a new tip for each transfer of matrix, and that quickly becomes a lot of tips, when it seems like 8 would do.
I am using 300 µL tips and transferring between 70 and 120 µL of matrix.
Does anyone have any liquid class settings or advice for this kind of task?

These are the setting i have been trying, along with a 3 x 300µL pre rinse, with not much luck. The amount of variation I am getting can be seen in the second image.

Thanks,
Ewan

Hi @ewanfyffe ,

Based on your parameters, I’m guessing this is a Jet Empty dispense. I have found that I get more consistent results for repeated aspiration/dispense when using a slower Surface Empty dispense and using the surrounding liquid to wick the droplets off the tip. Even if it is an empty well, you can dispense at 2 mm from the bottom with no liquid following to mimic this behavior as the volume that is being dispensed will surround the tip.

I would recommend trying a something like the following:

Hi Brandon,

You are correct, i have been using a jet empty. I got 100 uL working pretty well, then tried, and gave up, on getting 40 to work. Thank you for the suggestion, i will go and try those settings now.
