Venus 6 : Power Steps (Serial Dilution)

I am using MicroLab Power Steps in Venus 6.2 version. I am stuck at the moment trying the serial dilution power step. Is this dilution limited to only on sample? I have samples (64) in a plate and trying to take each column from sample plate and perform a serial dilution in a separate dilution plate. I added all the samples , diluent and dilution plates as sequence as part of the power step setup. When i run the method it just performs the serial dilution for the first column of samples and ends the method as complete. Am i missing anything? I confirmed that the Auto-increment is enabled for samples. No luck !

Hi @naveen ,

Sorry to see you are running into this issue. I was able to reproduce this on my end as well. This has been sent to our R&D team for investigation. It appears to be resetting the source sequence back to 1 each time and capping out the sequence for the first column which is why it stops at 1.

I was able to get around this by creating a temporary sequence for the current column before feeding that into the PowerStep and then looping that over the source sequence. See below for my example:

Question: Is there a way to use different dilution factor for each dilution in the power step for e.g. : use dilution 8 in the first dilution and 9 in the second dilution?

Hi @naveen ,

Yes, you can! If you click on the Options button and check the box for Different volumes for each dilution, this allows you to type in your individual dilution factors:

@BrandonBare_Hamilton Question: Can a power step handle NTR tips by taking out the empty rack? Is there any provision to include the move within Power steps? Thanks!

Hi @naveen ,

Unfortunately, no. Power Steps do not take into account if the tip rack is an FTR or NTR. You would have to handle the removal and ensure enough tips are on the top racks before entering the Power Step.
